For decades, scientists have explored the use of liposomes to deliver chemotherapy drugs to tumor cells. But drugs can sometimes leak out of liposomes before they reach their destination, reducing the dose received by the tumor and causing side effects in healthy tissues.
Now, researchers report “Nanobowl-Supported Liposomes Improve Drug Loading and Delivery” in the American Chemical Society’s Nano Letters a way to stabilize liposomes by embedding a stiff nanobowl in their inner cavity.
“Here, we develop a new method to enhance the stability of actively loaded liposomal doxorubicin (DOX) through embedding a stiff nanobowl in the liposomal water cavity. Nanobowl-supported liposomal DOX (DOX@NbLipo) resists the influence of plasma protein and blood flow shear force to prevent drug leakage. This approach yields improved drug delivery to tumor sites and enhanced antitumor efficacy. Compared to alternative methods of modifying liposome surface and composition for stability, this approach designs a physical support for an all-aqueous nanoliposomal cavity. Nanobowl stabilization of liposomes is a simple and effective method to improve carrier stability and drug delivery,” write the investigators.
Scientists have tried various approaches to prevent liposomes from leaking, such as coating their surfaces with polymers or crosslinking lipids in their bilayers. However, these modifications can alter the properties of liposomes so that they interact differently with cells. Chao Fang, PhD, and colleagues from the Shanghai Jiao Tong University School of Medicine, and Jonathan Lovell, PhD, from the State University of New York at Buffalo, wanted to find a new way to stabilize liposomes that keeps their surfaces intact.
